Bodily secretions, during and after sexual intercourse,
are deposited on various personal items. These items may include,
undergarments, clothes, condoms, feminine hygiene products, bed sheets,
towels, toilet/bathroom tissues, carpeting and mattress. These deposited
bodily fluids remain a valuable source as dried stains for many years.
Semen deposits contain prostatic acid phospatase (PAP) enzyme, Prostate
Specific Antigen (PSA) and millions of sperms. These markers are routinely
used to determine presence of semen in forensic samples. Dried
stains can be tested to determine the presence of PAP enzyme. However,
this test is highly Presumptive because many other body fluids, specially
vaginal secretions, also contain PAP enzymes. The presence of PAP in
vaginal secretions and other body fluids, makes this technique unreliable
for the confirmatory detection of sperm/semen and may lead to False
Positive Results. Also PAP enzyme in older stain samples
becomes inactive and can NOT be detected with this test and may provide False
Negative Results.
Sperm detection using DNA technology is the most powerful
technique. Even a small amount of male specific DNA can be successfully
detected using DNA based techniques. When
there is question of infidelity DNA testing designed
specifically to detect male specific Y
chromosome can
provide vital answers. The
DNA profile of the sperm that is detected by Y chromosome
analysis then can be matched or compared to other
samples or DNA from other sources. Sperm detection
services may provide unquestionable evidence for
infidelity. Sperm
